The Studies on Pharmacological Actions of 2(2.4.5 Trichlorophenoxy) Ethyl-N-triethyl Ammonium Bromide(TETB)

Abstract
2 (2. 4. 5 Trichlorophenoxy) ethyl-N-triethyl ammonium bromide (TETB) is a derivative of quaternary ammonium compound, and it is closely related chemically to tetraethylammonium bromide or xylocholine bromide.
The present report is concerned with general pharmacological properties, toxicity and actions on automonic nervous system of this drug.
1.) Toxic effect and general pharmacological action.
1. Toxic effect : Main toxic effects of TETB to mouse were motor excitment, clonic convulsion and depression of respiration. Its LD_(50) was 99§·£¯§¸ intraperitoneally.
2. On isolated frog¢¥s heart : It produced slight possitive inotropic effect In concentration of 10^(-2), A-V block and standstill ill concentration of 10^(-2).
3. On the vein of isolated rabbit¢¥s ear : It produced slight dilatation in concentration of 10^(-3) and temporary dilatation followed by constriction in concentration of 10^(-2).
4. On isolated rabbit¢¥s intestine : Tone was reduced in concentration of 10^(-4)~ 10^(-5) and tone and motility were depressed completely in concentration of 10^(-3).
5. On isolated nonpregnant uterus of rabbit : Rhythmicity was depressed in concentration of 10^(-4) or more.
6. On the blood pressure of rabbit and cat : It caused fall of blood pressure in dose from 0.5§·£¯§¸, intravenously and in dose of 5§·£¯§¸, fall of 80mm Hg of bIood pressure lasting about 30 minutes.
2.) Action of TETB on autonomic nervous system.
1. On isolated intestine of rabbit : It was shown that TETB has antimuscarinic property because it relaxed intestinal spasm induced by acetylcholine but not by Bacl©ü
2. It inhibited the pressure response produced by occlusion of common carotid artery, stimulation of vagus nerve and injection of nicotine, while it did not antagonize the pressor action induced by tyramine, ephedrine and noradrenaline.
3. The contractive response on nictitating membrane of the cat produced by stimulation of preganglionic fiber of the cervical sympathetic nerve was interfered completely with TETB, while it did not inhibit the contractive action induced by adreraline, ephedrine and noradrenaline.
4. The results of this experiment suggested that TETB would block the autonomic ganglion.
5. TETB accelerated the development of tachyphylaxis of tyramine.
